Formula
Apparent Volume of Distribution: Vd (L) = Dose (mg) ÷ C₀ (mg/L)
Vd (L/kg) = Vd (L) ÷ Body Weight (kg)
Loading Dose (mg) = [Vd (L) × Desired Target Peak (mg/L)] ÷ Bioavailability (F)Clinical Pearls
Determines the exact initial loading dose required for critical emergencies (septic shock antibiotics, antiarrhythmics) and guides extracorporeal toxin clearance.
Limitations
Vd is an "apparent" mathematical parameter, not a true anatomical volume, and can exceed 1,000 Liters for drugs with high intracellular tissue affinity.
